Description
Indole-3-Acetic Acid (IAA) is a natural auxin, a plant growth regulator or plant hormone that is required for the proper growth and development of plants.
IAA plays an important role in plant cell enlargement and division, tissue differentiation, and responses to light and gravity. In plant tissue culture, it is used to support organ development such as shoot and root formation and can be incorporated into growth media such as MS or Gamborg's media for callus initiation.

Assay Protocol
Follow the steps below to prepare 1 L of tissue culture media using IAA powder:
- Prepare liquid tissue culture media using PCT's MS, DKW, or another suitable alternative medium.
- Add sugar according to the quantity of media being prepared. Generally, 30 g of sugar is added per 1 L of medium.
- Add 1 mL/L PPM and 3-4 g/L gellan gum or 6 -8 g/L agar.
- Add the desired quantity of plant hormone from the prepared PGR stock solution.
- Autoclave the medium for 20 minutes at 15 psi and 121°C.
- The prepared tissue culture medium is ready for use.
